Can I use Weaning Mode with an increased Motion Start Level in the Happiest Baby App?

No, Weaning Mode can only be used when the baseline is at the default setting (Blue).

Since Weaning restricts movement on Baseline, it would be counterproductive to SNOO's benefits of having the Motion Start Level set higher. In fact, we recommend that parents who have SNOO starting on a higher motion level change it back to Blue Baseline two weeks prior to engaging Weaning Mode (to get the baby used to less and less motion).
